Guanfacine Hydrochloride 1mg, Oral tablet, extended release

Alcohol

· Severity: Major

· Notes for Consumers: Avoid alcohol and alcohol-containing products while taking Guanfacine. You may feel drowsy or more tired when taking these drugs together. Do not drive or operate machinery until you know how these drugs affect you. If you notice slurred speech, confusion, difficulty concentrating, dizziness, or fainting, contact your health care provider right away.

· Notes for Professionals: Advise patients to avoid use of alcohol while on guanfacine treatment. Guanfacine has been associated with sedative effects and can potentiate the actions of other central nervous system (CNS) depressants including alcohol. Before using guanfacine with other centrally active depressants consider the potential for additive sedative effects. Caution patients against operating heavy equipment or driving until they know how they respond to treatment with guanfacine.

Grapefruit juice

· Severity: Major

· Notes for Consumers: Do not take these agents together without approval from your prescriber. Taking these agents together could result in serious side effects such as dizziness; feeling faint or lightheaded, falls; unusually weak or tired; unusually slow heartbeat. Report any of these effects immediately should they occur.

· Notes for Professionals: Avoid consumption of grapefruit or grapefruit juice with guanfacine. Grapefruit is a strong CYP3A4 inhibitor, and guanfacine is a CYP3A4 substrate. Consumption may significantly increase exposure to guanfacine and increase the risk of alpha-adrenergic effects (e.g., hypotension, drowsiness, lethargy, bradycardia).
